Dancing cheek to cheek: Cryptococcus neoformans and phagocytes

چکیده

Meningoencephalitis caused by Cryptococcus neoformans (Cn) has become one of the leading causes of mortality in AIDS patients. Understanding the interactions between Cn and phagocytes is fundamental in exploring the pathogenicity of cryptococcal meningoencephalitis. Cn may be extracellular or contained in the monocytes, macrophages, neutrophils, dendritic cells and even endothelial cells. The internalized Cn may proliferate inside the host cells, or cause the lysis of host cells, or leave the host cells via non-lytic exocytosis, or even hijack the host cells (Trojan horse) for the brain dissemination, which are regulated by microbe factors and also immune molecules. Coexistence of protective and deleterious roles of phagocytes in the progression of cryptococcosis warrant further investigation.

Effect of mannose-binding protein on binding of Cryptococcus neoformans to human phagocytes.

The capacity of human peripheral blood monocytes, neutrophils, and monocyte-derived macrophages to bind the yeast Cryptococcus neoformans was increased when the phagocytes were cultured on surfaces containing recombinant human mannose-binding protein. In contrast, soluble mannose-binding protein had no effect on cryptococcal binding by phagocytes.

Effect of amphotericin B on capsule and cell size in Cryptococcus neoformans during murine infection.

Antifungal drugs can affect the cellular morphology of Cryptococcus neoformans in culture, which alters its interactions with phagocytes. We examined the effects of amphotericin B on C. neoformans during murine infection. The antifungal reduced capsule size and serum polysaccharide, which suggests an additional mechanism for amphotericin B's efficacy in cryptococcosis.
